Na, knowing how to hold a real guitar is the only thing that will actually help you on the game, the game has buttons not strings so IMO the only thing the two things have in common is:
They can both be held the same and they can both be plucked :D
No...in fact anyone with musical talent will find it hard to play at first because it's not about playing with the music but rather hitting the buttons at just the right time and that time is not with the beat usually.
An Experienced Guitar Hero player vs An Experienced Guitarist at GH the GH player will win.
But if both have never played the game before I think the Guitarist would win.

i feel that as a guitarist you gain no benefit over others. as a guitarist with good rhythm you have a great benefit over others. overall i think that if you play drums you're probably the best at guitar hero. the game is all about rhythm and coordination. I play guitar and drums, among other instruments, i should hope that i would know.

real guitars are played so differently than guitar hero that they would end up playing worse then a normal person who doesnt play guitar hero, on guitar hero 3 the guitarist that has the guitar "arm the homless" tried playing against his sons in a vid for the game and sucked but his sons did great
I play acoustic and electric and when I picked up GH, it was very hard and it took me some time to adjust. I think the hardest part was just adjusting to the fact that you had to stare at the screen more and not concentrate on the buttons.
However, I have now been able to play expert and hard, so I'm a pretty decent player now. But I disagree with what a guy said above; if two people were going to play GH for the 1st time and a guitar player was one of those two, the guitar player would be worse, just trust me on it.
